Understanding the Toxicity and Repurposing Potential of Kinase Inhibitors

Abstract
Current FDA approved kinase inhibitors have significant side-effects, many of which might be due to the off-targets of these drugs. Dr. Hammad Naveed will present a novel computational approach for large-scale discovery of new targets for existing drugs. For a given drug, he will construct the 3D signature of drug binding sites that capture its promiscuous structural features. He is able to predict the interaction of six FDA approved kinase inhibitors with its known targets with a sensitivity of 50% and specificity of 56%. 4 out of the 5 top predicted targets were shown to interact with Sorafenib using in vitro experiments.
